肾必宁冲剂对肾小球系膜细胞凋亡及bcl-2、ICE基因表达的影响
Influence of "Kidney Peace Granules" on Apoptosis of Mesangium Cell and bcl-2 and ICE Genetic Expression
【摘要】 为探讨肾必宁冲剂对肾小球系膜细胞凋亡及凋亡相关基因ICE和Bcl 2表达的影响 ,阐明治疗系膜增生性肾病的机制 ,35只大鼠随机分为空白组 (A)、中药高剂量组 (B)、中药中剂量组 (C)、中药低剂量组 (D)、环磷酰胺 (CTX)组(E) ,每组各 7只 ,分别予生理盐水 (A组 )、肾必宁冲剂 (B、C、D组 )、CTX胃饲 ,8d后分别处死取血清 ,采用肾小球系膜细胞培养方法进行体外实验研究 ,测定系膜细胞增殖指数 ,观察凋亡细胞核小体DNA的断裂现象 ,并检测其ICE和bcl 2的变化。结果 :加入肾必宁冲剂血清 16h后 ,系膜细胞出现了典型的凋亡征象 ,凋亡率显著高于A组(P <0 .0 1) ;B、C、D组ICE表达明显高于A组 (P <0 .0 1) ,而bcl 2基因各组均表达不明显 (P >0 .0 5 )。提示肾必宁冲剂可诱导系膜细胞凋亡 ,促进肾小球增殖性病变消散 ,并可能通过调控ICE和bcl 2基因影响凋亡
【Abstract】 In order to explore influence of"Kidney Peace Granules"on death of mesangium cell and the related ICE and Bcl 2 genetic expression and to explain the therapeutic mechanism of nephropathy due to mesangial proliferation,35 rats were randomly divided into the control group(Group A),high dose group of Chinese medicine(Group B),medium dose group of Chinese medicine(Group C),low dose group of Chinese medicine(Group D)and cytoxan(TCX) group(Group E),7 rats each group,and were respectively fed physiological saline(Group A),"Kidney Peace Granules"(Group B,C and D)and CTX,for respective management of serum after 8 days,by external experimental study of mesangium cell culture,to determine the proliferation indexes of the mesangium cell and observe the dying cell and breaking phenomenon of nucleosome DNA and determine ICE and bcl 2 changes.The results showed that in serum 16 hours after"Kidney Peace Granules"was used,the typical dying sign of mesangium cell appeared,with the dying rate remarkably higher than Group A( P < 0.01 )and ICE expression obviously higher in Group B,C and D than in Group A( P < 0.01 )and without obvious bcl 2 genetic expression in various groups( P > 0.05 ).These findings indicate that"Kidney Peace Granules"can induce the death of mesangium cell,promote dispersion of the proliferating change of the glomerulus and may influence the death by regulating ICE and bcl 2genes.
