β-榄香烯对大鼠胃癌及癌前病变DNA含量、增殖细胞核抗原表达及端粒酶活性的影响

【摘要】 目的 研究β 榄香烯对癌变大鼠胃黏膜端粒酶活性、增殖细胞核抗原表达及DNA含量的影响。 方法 采用MNNG、10 %NaCl混合液及酒精灌胃建立大鼠胃癌模型 ,设立正常空白组、模型组及 β 榄香烯组 ,分别采用免疫组化、Feulgen染色及端粒重复扩增微孔板杂交法检测大鼠胃黏膜增殖细胞核抗原表达、DNA含量及端粒酶活性。结果 β 榄香烯组癌变发生率、DNA含量、增殖细胞核抗原表达及端粒酶活性均低于模型组。在大鼠癌变过程中 ,随着胃黏膜癌变的发展 ,DNA含量、增殖细胞核抗原表达及端粒酶活性均呈递增趋势 ,且两者与端粒酶活性具有重要的相关性。结论 β 榄香烯抗癌 ,抑制端粒酶活性是其重要机制之一。

【Abstract】 Purpose To study the effects of β elemene on DNA content,proliferating cell nuclear antigen(PCNA) expression and telomerase activity of gastric cancer in Wistar rats. Methods We studied DNA content,expression of PCNA and telomerase activity by using feulgen,TRAP microtiter plate hybridization assay in three groups of rats:normal control,model control,treatment groups with β elemene in gastric cancer rats induced by MNNG,40% alcohol and 10% sodium chloride. Results Compared with elemene group,the rates of gastric cancer,DNA content,PCNA expression and telomerase telomerase would be lower than ones of model ones.With the development of gastric cancer,the expression of PCNA and DNA content and telomerase activity were increased by degress,and the former has an important correlation with the latter. Conclusions β elemene can repress telomerase activity of gastric cancer.
