深Ⅱ度烧伤创面细胞bFGF和PDGF对愈合的影响

【摘要】 目的研究bFGF和PDGF在深Ⅱ度烧伤愈合中的表达特点 ,以及与创面愈合中的关系。为临床合理应用外源性的bFGF和PDGF提供实验依据。 方法采用地高辛细胞原位杂交方法检测SD大鼠深Ⅱ度烫伤模型在伤后不同时段的创面渗出液细胞的bFGF和PDGF中mRNA表达变化。 结果实验组的bFGF是在 5d开始表达 ,在 7d达到高峰 ,然后缓慢下降 ,至 14d时还有一定水平的表达 ,对照组bFGF的表达水平都较低 ,仅在 5d有低水平表达 ,14d已基本无表达。在检测PDGF时 ,实验组 1~ 3d处于较低水平表达 ,7d开始高水平表达 ,10d是表达的高峰 ,随后下降。对照组PDGF的表达在 1~ 3d处于较低水平 ,5~ 14d有一个低水平的mRNA表达延迟峰段。 结论创面渗出液细胞bFGF、PDGF的mRNA表达的启动、表达的高峰和减弱与深Ⅱ度烧伤的创面愈合状况有关 ,与相关细胞在创面聚集、诱导和分泌bFGF、PDGF有关。

【Abstract】 Objective To study the characteristics of the expression of bFGF and PDGF in deep Ⅱ 0 burn wound healing and their relationship to burn wound healing, so as to provide evidence to the clinical application of exogenous bFGF and PDGF. With SD rat burn models, we detected the change of expression of bFGF and PDGF mRNA in the exuded cells from superficial and deep Ⅱ 0 burn wounds by DIG in situ hybridization at different times. bFGF in the treated groups started to express on 5th day and reached the top on 7th day, then declined slowly, but still expressed on 14th day. In contrast, bFGF in the control groups expressed in low levels, rather low on 5th day and not at all on 14th day later, while PDGF, the treated groups expressed in low levels from 1st to 3rd day, started to express in high level on 7th day, reached the peak on 10th day and then dropped. PDGF expression in the control groups was in low levels during 1st to 3rd day, and there was a low expression mRNA on 5th to 14d. The study showed that bFGF\, PDGF mRNA expression promotion, as well as then expression peak and decline in exuded cells from burn wounds is related to deep Ⅱ 0 burn wound healing and also to the correlated cells gathering in the wounds, inducing and secreting bFGF, PDGF.
