小鼠腔前卵泡的分离采集和体外成熟研究
Isolation and in Vitro Maturation of Mouse Preantral Follicles
【摘要】 采用机械法和酶法结合机械法分离采集小鼠卵巢 ,每只小鼠可获腔前卵泡 5 3 2 5和 5 7 2 9枚 (P <0 0 5 )。成年小鼠腔前卵泡在体外培养 9d ,换用成熟培养液继续培养 2 4~ 2 8h后 ,3种不同培养系统中卵泡的成熟分裂率分别为 5 47%、3 85 %和 9 31%。表明EMEM +5 %FCS +0 2 3mmol/mL丙酮酸钠 +2 0 μg/mLFSH +0 0 5mmol/mL次黄嘌呤 +2 0 0IU/mL青 -链霉素培养系统可以使小鼠腔前卵泡卵母细胞在体外发育达到成熟
【Abstract】 In present experiment, the preantr8l follicles are isolated from mouse ovaries by mechanical and enzyme digestion combined with mechanical treatments. The average number of preantral follicles, collected by two treatments, are 53 25 and 57 29 preamral follicles per pair of ovaries (P<0 05). Maturation rate of three culture systems are 5 47%, 3 85% and 9 31% for 9 days culture. There are significant differences among the three culture systems(P<0 05). It appears that EMEM+5% FCS+0 23mmol/mL sodium pyruate+20μg/mL FSH+0 05mmol/mL Hypoxahine+200IU/mL penicillin streptomycin is suitable for in vitro maturation mouse preanral follicles.
