叶绿素缺乏大麦突变体叶绿体结构功能及生化特性的研究
STUDIES ON THE STRUCTURE AND FUNCTION OF CHLOROPLAST IN A CHLOROPHYLL-LESS BARLEY MUTANT(NYB)
【摘要】 突变大麦类囊体膜吸收光谱的蓝区峰值明显高于野生型 ;A683 /A652 比值较高 ;CD谱信号较弱 ,这说明其捕光色素蛋白复合物发生了变化 .SDS PAGE结果表明 ,突变大麦的Rubisco的水平与野生型一致 ,但在类囊体膜上分子量为 2 4~ 30kD的LHCⅡ蛋白组分减少
【Abstract】 A comparative study of chloroplast ultrastructure, absorption spectrum and SDS-PAGE analysis was made in a chlorophyll-less barley mutant and its wildtype. The results showed that the mutant barley has lower Chl and carotenoid contents but higher Chl a/b ratio.NYB chloroplast possesses less thylakoids and grana stacks.As compared to the wild type barley, the light absorption of NYB thylakoids was much higher within 450~480 nm and 600~640 nm.SDS-PAGE results indicated that there is no differences in Rubisco level but a bigger decrease in LHCⅡ apoproteins between mutant and its wildtype.
