甜瓜ACC氧化酶前导激应元件的克隆与序列分析

【摘要】 以甜瓜基因组DNA为模板,利用PCR技术扩增获得甜瓜ACC氧化酶基因家族成员CM-ACO1的前导顺式元件,长度为 886bp。将其克隆至pGEM-T easy载体上。序列分析结果表明:克隆的 CM-ACO1基因顺式元件的核苷酸序列与已报道的相比有99.6%同源,元件中含BRE,ERE,WUN等,能接受多种外界信号刺激。

【Abstract】 An 866 bp fragment of the 5’ non-translated region of ACC oxidase gene CM-ACO1 was amplified by PCR using total genomic DNA from melon as template. The amplified fragment was cloned into the plasmid of pGEM T easy and sequenced. The resultant nucleotide sequence had a homology of 99 % as compared with the reported sequence. Motifs of BRE, ERE, WUN with significant homology to known cis-acting elements were identified in the cloned nucleotide sequence. Analysis revealed that these elements were in response to various stress.
