水稻受稻瘟病菌诱导的转座类似蛋白新基因RIM2的分离与鉴定

【摘要】 以近等基因系H7R和H7S为材料,运用差别显示(DD-PCR)技术获得RMa19片段。用此片段为探针筛选c DNA文库,获得 3 944 bp全长基因 RIM2。Northern杂交结果显示该基因在水稻抗感品系中均受稻瘟病菌的强烈诱导。RIM2 ORF编码 653个氨基酸,其序列与多种植物转座类似蛋白有 32%~55%的同源性。其 5'端约 800 bp与 Xa21基因家族A1,C的非编码区有82%的序列同源性。Southern杂交表明RIM2多拷贝存在与水稻基因组中。RIM2为一新的受病菌诱导的早期反应基因。

【Abstract】 A rice gene RIM2 was identified by mRNA differential display, it was strongly induced by both incompatible and com- patible strains of Magnaporthe grisea. A 3.9 kb cDNA clone was identified and predicted to encode a 653 amino-acid protein, which is 32%~55% identical with proteins encoded by the CACTA transposons of other plants.AT-rich region was found in the 5’ and 3’ noncoding termini of RIM. Sequence of RIM2 was 82% identical with sequences flanking the Xa21 gene family members, Al and C, the disease resistance genes. Southern blot hybridization with genomic DNA from different rice isolines indicated that RIM2 is present in multiple copies. These results suggest that RIM2 might be involved in the early rice response to infection induced by the fungal pathogen.
