欧文氏菌AEBL002菌株八氢番茄红素脱氢酶基因保守序列的克隆与同源性分析
COLONING OF CONSERVATIVE SEQUENCE OF PHYTOENE DESATURASE GENE FROM AEBL002 STRAIN AND ITS HOMOLOGY ANALYSIS
【摘要】 从农田土壤中分离筛选到一株产胡萝卜素菌 AEBL0 0 2 ,提取其基因组 DNA,根据报告的八氢番茄红素脱氢酶基因设计 PCR引物 ,对 AEBL0 0 2基因组 DNA进行 RCR扩增 .扩增产物为一条大小约为 80 0 bp的 DNA片段 ,在 Genbank中对其进行同源性检索 ,发现该片段与 Erwinia uredovora八氢番茄红素脱氢酶基因在 73 2个碱基中有 96.0 3 8%的一致性 .
【Abstract】 A strain which can produce carotenoid was screened from soil in farm. Genomic DNA was isoated from this strain and PCR was done to amplify conservative sequence of crt Ⅰ gene encoding phytoene desaturase. The product of PCR reaction is a 800bp DNA fragment. This fragment display sequence identities of 96.038% with phytoene desaturase gene of E.uredovora in Genbank.
