藻胆蛋白与单克隆抗体交联试剂的探索
Research on reagents of phycobiliprotein conjugating with IgG
【摘要】 目的选取浓度适宜的异双功能试剂用于藻胆蛋白荧光标记物的制备。方法选用 SPDP、SMCC及 2 - IT分别对 R-藻红蛋白和抗乙肝病毒表面抗原单克隆抗体进行处理 ,研究了不同浓度异双功能试剂对 R- PE荧光特性和 Ig G活性的影响。结果从保持 R- PE荧光特性和 Ig G活性的角度 ,异双功能试剂在修饰 R- PE或 Ig G时存在一个最佳浓度 :SPDP摩尔浓度小于 1.6 6 7m mol/ L ,SMCC摩尔浓度小于 0 .6 6 7m mol/ L ,2 - IT摩尔浓度小于 12 .5 mm ol/ L较适宜。结论须进一步考证异双功能试剂浓度对交联效率及交联物质量的影响以确定最佳的异双功能试剂组合及浓度。
【Abstract】 ObjectiveTo choose proper conditions to develop phycobiliprotein conjugated IgG. MethodsR-phycoerythrin and anti-HBsAg served as models. The effects of heterobifunctional reagents with different concentrations on R-phycoerythrin and IgG were studied.Results It is suitable for both R-phycoerythrin and IgG that the concentration of SPDP is not more than 1.667 mmol/L,SMCC not more than 0.667 mmol/L,and 2-IT not more than 12.5 mmol/L. Conclusion Many other factors should be investigated to get the best conjugating conditions for phycobiliprotein conjugated IgG. [
