R-藻红蛋白固相免疫荧光检测中洗涤条件的筛选

【摘要】 目的筛选 R-藻红蛋白免疫荧光检测中适宜的洗涤条件。方法采用二步法改变洗涤液离子强度、p H值 ,比较弱阳性血清的检出率及阴性假阳性率 ,由此筛选最佳洗涤条件。结果洗涤液离子强度的增加有助于降低阴性血清假阳性现象。在本实验所涉及样品范围内 ,摩尔浓度大于 10 0 mm ol/ L 时 ,假阳性率可降低到 0 ;12 0 m mol/ L 时 ,阳性检出率最高值达93.3% ;中性、偏碱性的 p H值对检出率影响较小。结论以 12 0 m mol/ L PBS(p H7.5 )为洗涤液有利于提高 R-藻红蛋白荧光免疫法的特异性和灵敏度。

【Abstract】 ObjectiveTo screen for the optimal washing buffer in R-phycoerythrin fluorescence immunoassay MethodsChange the ionic intensity and pH value of the washing buffer. Screen the washing buffer by comparing the checkout ratio of positive samples and the pseudo-positive ratio of negative samples.ResultsIncreasing ionic intensity can be helpful to decrease pseudopositive phenomenon in negative samples and to increase sensitivity when concentration is below 120 mmol/L. Pseudopositive ratio can be decreased to zero. when concentration is near to 120 mmol/L and the positive checkout ratio can be up to 93.3%. But much higher concentrations may restrict the sensitivity to be increased. A weak basic washing condition is appropriate.Conclusion120 mmol/L PBS (pH7.5) is suitable for R-phycoerythrin fluorescence immunoassay both in the diagnosis specificity and in the sensitivity. [
