镉雌激素样作用的实验研究
Experimental Study on the Estrogen like-Effect of Cadmium
【摘要】 [目的 ]评价镉的雌激素样效应。[方法 ]从体外和体内两个水平 ,观察不同浓度的氯化镉对MCF 7人乳腺癌细胞生长和去卵巢SD大鼠子宫的诱导增生作用。 [结果 ] 10 -6mol/L的氯化镉使MCF 7细胞增殖达最大 ,为对照的3 92倍 ,并且其诱导增殖作用被纯抗雌激素ICI 182 ,780完全阻断。镉的相对增殖效率为雌二醇的 6 5 % ,相对增殖能力为雌二醇的 1/ 10 0 0。每天 0 1、0 5及 2 5mg/kg连续 3d腹腔注射氯化镉能明显促进去卵巢大鼠子宫的增生 ,并且具有剂量 效应关系。 [结论 ]镉能够模拟雌二醇诱导MCF 7细胞增殖及大鼠子宫增重。镉具有雌激素样效应。
【Abstract】 Objective] The aim of this study was to evaluate the potential estrogenic effect of cadmium. [Method] Both in vitro (proliferation of MCF 7 human breast cancer cell) and in vivo (uterotrophic assay in ovariectomized SD rats) assays were used in this study. [Results] 10 -6 M cadmium chloride induced the maximal cell proliferation of MCF 7 cells up to 3 92 fold compared to control group, and this effect was blocked completely by a pure antiestrogen ICI 182,780. The relative rate of the proliferation effect of cadmium was 65%, and the relative proliferation potency was 1/1000.0 1,0 5 and 2 5mg/kg per day by intraperitoneal injection of cadmium chloride for 3 days increased the weight of uterus of ovariectomized SD rats significantly in a dose related pattern. [Conclusion] It was suggested that cadmium can mimic the effect of estrogen to stimulate the growth of MCF 7 cells and the uterus of ovariectomized SD rats.Cadmium had estrogen like effect.
