内皮素-1和诱生型一氧化氮基因mRNA在肝硬变门静脉高压症大鼠肝组织中的表达

【摘要】 目的 :检测肝硬变门静脉高压症 (CPH)大鼠肝组织中内皮素 - 1(ET - 1)和诱导型一氧化氮合成酶 (iNOS)的表达情况 ,研究两者与肝内微循环改变和门静脉高压形成的关系。方法 :应用逆转录和多聚酶连反应 (RT -PCR)的方法测定CCl4 诱导CPH大鼠肝组织中ET - 1和iNOS基因mRNA的表达。结果 :ET - 1基因mRNA在CPH大鼠肝组织中的表达明显高于对照组 (增高 5 0 %± 13% ,P <0 .0 0 1) ,而iNOS基因mRNA的表达比对照组轻度减弱 (减弱 15 %± 8% ,P >0 .0 5 )。结论 :CPH大鼠肝组织中ET - 1表达增强、iNOS表达减弱与肝硬变时门静脉阻力增高有关。

【Abstract】 Objective: Endothelin-1 and NO could be involved in the regulation of hepatic microcirculation and in the development of portal hypertension. The expression of ET-1 and iNOS in normal and cirrhotic liver tissue were investigated. Methods: ET-1 and iNOS mRNA in liver tissue from normal, and cirrhotic rats with portal hypertension by repeated CCl 4 administration, were analyzed using reverse transcription-polymerase chain reaction. Results: ET-1 expression was markedly overexpression(50%±13% increased over control, P <0.001), but iNOS was reduced(15%±8% decreased than control, P >0.05) in the liver tissue after CCl 4-induced cirrhosis with portal hypertension. Conclusion: Overexpressed ET-1 and reduced iNOS by cirrhotic rat liver, may contribute to the increased resistance to portal flow in cirrhotic liver.
