PCR-SSCP方法研究血液系统肿瘤的p53基因突变
Detection of p53 gene mutations in hematologic malignancies by PCR SSCP analysis
【摘要】 目的 :探讨 p5 3基因突变在血液系统肿瘤发病及进展中的作用。方法 :采用 PCR- SSCP方法对 34例血液系统肿瘤患者 p5 3基因第 5、8外显子进行研究。结果 :从 1例急性粒细胞白血病 (AML ) (1/ 12 ,8.33% ) ,1例急性淋巴细胞白血病 (AL L) (1/ 5 ,2 0 % ) ,2例非霍奇金淋巴瘤 (NHL) (2 / 5 ,40 % ) ,2例多发性骨髓瘤 (MM) (2 / 8,2 5 % )中发现了 p5 3基因突变。其中 1例 AML,1例 NHL 及 1例 MM,均在检测后 3个月内死亡。结论 :p5 3基因突变在血液系统肿瘤的发病及疾病发展中起着一定的作用
【Abstract】 Objective:To discuss the role of p53 gene mutations in the pathogenesis of hematologic malignancies.Method:Exon 5、8 of the p53 gene were scanned in 34 cases of hematologic malignancies by PCR SSCP.Result:p53 gene mutations were detected in 1 of 12( 8.33 %)patients with acute myelogenous leukemia(AML),1 of 5(20%) with acute lymphoblastic leukemia(ALL),2 of 5 (40%)with non Hodgkin′s lymphoma(NHL) and 2 of 8 (25%) with multiple myeloma( MM),in which one case of AML,one case of NHL and one case of MM died in three months after detection.Conclusion:The p53 gene mutations plays a certain role in the pathogenesis and progression of hematologic malignancies.
