补肾活血泄浊汤对肾小球上皮细胞蛋白质和硫酸肝素蛋白多糖合成的影响
Effects of Bushenhuoxuexiezhuo Decoction on Syntheses of Protein and Heparin Sulfate Proteoglycan in the Cultured Glomerular Epithelial Cell of Rat
【摘要】 探讨脂多糖 (LPS)、硫酸鱼精蛋白及补肾活血泄浊汤对体外培养大鼠肾小球上皮细胞(GEC)蛋白质和硫酸肝素蛋白多糖 (HSPG)合成的影响。应用LPS、硫酸鱼精蛋白作为刺激因子 ,观察其对GEC蛋白质和HSPG合成的影响 ;并采用血清药理学方法 ,提取动物的含药血清作用于上述受刺激的上皮细胞 ,观察补肾活血泄浊汤对上皮细胞蛋白质和HSPG合成的影响。LPS和硫酸鱼精蛋白具有抑制GEC合成蛋白质及含硫化合物的作用 ;而补肾活血泄浊汤可显著拮抗上述因子的抑制细胞蛋白质和HSPG合成的作用。肾小球上皮细胞是补肾活血泄浊汤发挥治疗作用的重要靶细胞 ,这可能是该方防治以蛋白尿为主要临床表现的肾小球疾病的机制之一。
【Abstract】 Effects of lipopolysaccharide (LPS), protamine sulfate (PS) and Bushenhuoxuexiezhuo Decoction (BD) on the syntheses of protein and heparin sulfate proteoglycan (HSPG) were studied in the cultured glomerular epithelial cell (GEC) of rat, by serum pharmacological method. The results showed that LPS and PS could markedly inhibited the syntheses of protein and sulfur compounds in the cultured GEC of rat, while BD could significantly antagonized the effects of LPS and PS. The results suggest that GEC is important target cell of BD, and exerting effects on GEC may be one of the mechanisms of BD in treating glomerulopathy with proteinuria as the main symptom.
