【摘要】 目的 探讨穿琥宁抑制甲3型（H3N2）流感病毒诱导细胞凋亡的研究。 方法 采用观察细胞形态学变化、计算调亡细胞的百分率和流式细胞仪测定等方法。 结果 ⑴在光镜下观察了流感病毒感染的狗肾传代细胞（MDCK），感染的MDCK细胞可见细胞凋亡特有的形态学变化。如：可见广泛的染色质凝集成数个紧密的块状、胞浆浓缩及胞浆内大量空泡形成等。⑵病毒感染组MDCK细胞凋亡的百分率显著高于其它对照组（P＜0．001）。⑶药物病毒组、药物对照组和细胞对照组比较无显著性差异（P＞0．05）。⑷抗流感病毒药穿琥宁（中草药）能明显抑制甲3型流感病毒诱导的细胞凋亡（P＜0．001）。⑸病毒感染组的MDCK细胞“亚G1峰”明显高于其它对照组。 结论 本研究说明甲型流感病毒能诱导细胞凋亡，而抗流感病毒中草药穿琥宁能抑制其诱导的细胞凋亡。更多还原

【Abstract】 Objective Explore the efficacy of chuanhuning through observing it inhibiting the apoptosis induced by influenza virus A3． Methods The morphological changes of MDCK cells infected by influenza virus were observed and the percentage of apoptotic cells were calculated by light microscope and flow cytometry． Results ⑴The morphological of Madin－Darby canine kidney cells(MDCK) infected with influenza virus A3 was observed under the light microscope． It was seen the infected cells with characteristic morphological changes of apoptosis such as extensive condensation of chromatin into dense masses concentration of cytoplasm and cytoplasm vacuolization． ⑵The effects of antiviral drugs on the apoptosis of the infected cells were analyzed． The results were shown as 1)The percentage of apoptotic MDCK in the virus infected group was significantly higher than the noninfected group(P＜0．001) 2)The percentage of apoptotic cells among three groups the virus infected cells treated with antiviral drugs the cells treated with antiviral drugs and cells had no significant differences(P＞0．05)； 3)It was observed that the antiviral drugs chuanhuning could significantly inhibit apoptosis induced by influenza virus A3(P＜0．001) 4)The sub－G1 peak in the group of infected MDCK was significantly higher than other group． Conclusion This study shows that influenza virus A3 can induce apoptosis and Chinese traditional herb Chuanhuning as anti influenza virus can inhibit the apoptosis induced by influenza virus A3．更多还原
