【摘要】 目的 :研究重组人白细胞介素 13 (rhIL 13 )对巨核细胞系 -Dami细胞原癌基因c mpl表达的影响。方法 :实验组细胞培养系统中分别加入 2 5ng/ml,5 0ng/ml,10 0ng/mlrhIL 13 ,对照组不加任何细胞因子。用RT PCR法检测c mplmRNA的表达。结果 :经 2 5ng/ml、5 0ng/ml、10 0ng/mlrhIL 13处理过的实验组Dami细胞较之对照组其c mplmRNA的表达分别提高 2 4 .8%、2 7.9%和 3 1.5 %。结论 :rhIL 13增强了Dami细胞原癌基因c mpl表达 ;rhIL 13促进Dami细胞增殖的部分机制可能是通过上调c mpl的表达来实现的。更多还原

【Abstract】 Objective: To investigate the effect of recombinant human interleukin 13(rhIL 13) on the expression of proto oncogene c mpl in Dami Cells, a human megakaryobiastic leukemia cell line. Methods:In experimental groups,the concentrations of rhIL 13 were 25 ng/ml, 50 ng/ml,and 100ng/ml. In control group, the concentration of rhIL 13 was 0 ng/ml. The expression of c mpl mRNA in Dami Cells was determined with RT PCR. Results:The expressions of c mpl mRNAs in groups of 25 ng/ml, 50 ng/ml, and 100ng/ml increased by 24.8%?27.9%, and 31.5%, respectively. Conclusion:The data indicate that rhIL 13 enhances the expression of proto oncogene c mpl in Dami Cells. Moreover, the stimulation of Dami Cells by rhIL 13 may be partly resulted from an increase of the proto oncogene c mpl gene expression.更多还原
