羟乙基淀粉对大鼠蛋白尿的治疗作用

【摘要】 目的 :探寻羟乙基淀粉治疗大鼠蛋白尿的可行性。方法 :建立蛋白尿大鼠动物模型 ,在静脉点滴 6 %羟乙基淀粉前后 ,通过大鼠输尿管插管收集肾后膀胱前尿液 ,测定尿蛋白分子量并比较治疗前后尿蛋白含量 ,采用聚丙烯酰胺凝胶电泳法对尿蛋白分子量进行分析 ,同时观察治疗前后血肌酐和尿素氮变化。结果 :羟乙基淀粉治疗后尿蛋白定量较治疗前明显下降 ,尿蛋白电泳表明高、中分子蛋白条带数目和含量较治疗前显著减少。治疗前后尿量、血肌酐和尿素氮无明显变化。结论 :羟乙基淀粉能够安全、有效地减少实验大鼠的蛋白尿 ,为探寻蛋白尿的新的治疗方法提供了理论依据

【Abstract】 Objective:To investigate the effect of Hydroxyethy1 Starch(HES) on proteinuria in rats induced by Daunorubicin. Methods: Proteinuria was induced with a single intravenous dose of Daunorubicin. 6% Hydroxyethy1 Starch was infused intravenously to each rat. Urine was collected from the ureter before and after treatment. The amount of urinary protein was measured by method of Coomassie brilliant blue stain. The molecular weight of urinary protein were analyzed by polyacrylamide gelelectropheresis(PAGE). Serum creatinine and blood urea nitrogen were also measured. Results: The amount of urinary protein decreased from (5.73±2.10)mg/h before treatment to (1.80±0.47)mg/h after treatment. The number of bands and amount of protein with greater molecular weight significantly decreased. Serum creatinine and urea nitrogen did not change significantly. Conclusion: Hydroxyethyl Starch is a safe and effective treatment as a leakage stopper on glomerular proteinuria.
