【Abstract】 Several definitions of Tm are discussed in the paper;(1)Tm is the temperature at which the hyperchromic effect of the DNA molecules heated to denature has increased to 50%; (2)Tm is the temperature at which half of the heated DNA molecules have denatured; (3)Tm is the temperature at which the absorption of UV light of the DNA molecules heated to denature has increased to half of the highest absorption of UV light.
