氟虫腈半抗原设计及抗体制备
Hapten Design and Antibody Preparation of Fipronil
【摘要】 以氟虫腈 (F)及其一种类似物 (FH)作为半抗原 ,采用碳化二亚胺法 (EDC)和戊二醛法将氟虫腈偶联到载体蛋白上 ,采用活性酯法将FH偶联到载体蛋白BSA和OVA上作为人工抗原。载体为BSA的偶联物作为免疫原免疫新西兰大白兔 ,其中FH2 934、FH2 962、F2 895产生了特异性抗体 ,间接ELISA测定效价分别为 2 5 60 0、12 80 0、1280 0 ,其中检测低限 (LLQ)最低达 0 2 μg/L。
【Abstract】 Fipronil(F) and one of its analogy(FH) were adopted as haptens.The former were coupled to carrier protein BSA and OVA by means of EDC and GD,and the later was coupled to carrier protein BSA and OVA by means of active ester These protein hapten conjugates were acted as man made antigens.New zealand rabbits were immunized by the antigen with the carrier protein of BSA.The results indicated that FH2934,FH2962 and F2895 elicited special antibodies to fipronil,and the titers are 25 600,12 800 and 12 800 respectively measured by indirect ELISA.The most low limited quality was 0 2 μg/L
