银杏叶提取物对缺氧复氧诱导大鼠大脑皮层神经细胞凋亡的保护作用

【摘要】 应用原代分离培养的Wistar胎鼠大脑皮层神经细胞缺氧复氧模型 ,研究银杏叶提取物对神经细胞凋亡的抑制作用。方法 :采用流式细胞术检测DNA含量及凋亡细胞百分率 ;原位末端标记法检测DNA断裂 ;光镜及透射电镜观察细胞形态学变化。结果 :银杏叶提取物能改善神经细胞亚微结构的变化 ,减少凋亡细胞数 ,使缺氧复氧诱导神经细胞凋亡百分率明显下降。结论 :银杏叶提取物对缺氧复氧所引起的胎鼠大脑皮层神经细胞凋亡有明显抑制作用。

【Abstract】 Objective:To explore the inhibition effect of extract of ginkgo biloba (EGB) on the neurons apoptosis induced by hypoxia/reoxygenation.Methods:The hypoxia/reoxygenation model primary culture of E15~17 day fetal rat cortical neurons was used.The DNA content and the percentage of apoptosis were measured by flow cytometry;DNA fragmentation was measured by nick end labeling (TUNEL) method;Morphological changes were observed with light microscope and electron microscope.Results:The result showed that hypoxia/reoxygenation medium induced apoptosis in primary cultured cortical neurons.EGB decreased the percentage of neuronal apoptosis,prevented DNA fragmentation and morphological changes of cortical neurons,and reduced the numbers of positive cells.Conclusion:The results indicate that EGB has obvious inhibition effect on the apoptosis of rat cortical neurons induced by hypoxia/reoxygenation.
