MPTP损伤侧猴中脑黑质组织的基因表达
A STUDY ON DIFFERENTIALLY\|EXPRESSED GENES IN THE SUBSTANTIA NIGRA FROM A MPTP\|INDUCED MONKEY MODEL OF PARKINSON DISEASE
【摘要】 目的 采用mRNA差异显示技术 (mRNAdifferentialdisplay,mRNADD) ,寻找MPTP诱导和未诱导的猴中脑黑质组织差异表达的cDNA。 方法 对诱导侧及未诱导侧的黑质mRNA用 3′端 3个锚定引物分别与 5’端 30种随机引物进行逆转录 聚合酶链反应 (RT PCR) ,扩增产物经聚丙烯酰胺凝胶电泳及硝酸银染色显示目的条带。结果 获得一批表达差异的cDNA片段 ,对其克隆后作DNA序列测定、同源性比较分析发现 :其中 2个克隆为未知序列 ,已被GenBank接收 ,接收号为AF15 916 1、AF15 916 2。 结论 MPTP诱导后黑质可能有新基因的表达
【Abstract】 Objective mRNA differential display(mRNA DD RT\|PCR) was used to study differentially\|expressed gene in bilateral substantia nigra(SN) from 1\|methy\|4\|phenyl\|1,2,3,6\|tetrahydropyridine(MPTP)\|induced monkey model of Parkinson disease(PD). Methods The bilateral substantia nigra mRNA was reversely transcribed and PCR\|amplified by using 3 docking primers and 30 random primers from unilateral monkey model of Parkinson disease; the amplification products were analysized by polyacryamide gel electrophoresis and AgNO 3 staining. Results Some gene expressions in MPTP\|injected SN are different from those in contralateral side of SN. Sequencing and homologous comparison are carried out for those genes, among which two are unpublished fragments. The sequences were registered by GenBank and the accession number is AF159161,AF159162. Conclusion Some novel genes are probably expressed in the substatia nigra after MPTP induction. The further affirmation is in progress.\;[
