大鼠脊髓神经元原代细胞的分散培养
Dispersion Culture of Primary Neurons in Rat Spinal Cord
【摘要】 目的 建立一种简单、易行、满足基本科研需要的脊髓神经元原代细胞分散培养法。方法 取Wistar大鼠胎鼠或新生鼠的脊髓 ,以胰蛋白酶消化和机械吹打相结合 ,将组织分散成细胞悬液 ,培养于含10 %胎牛血清和阿糖胞苷 (抑制胶质细胞的增殖 )DMEM培养基中 ,观察脊髓神经元细胞生长状况。结果 3~ 6h细胞开始贴壁 ,胞体周围渐有突起 ;2~ 3d胞体明显增大 ,突起生长旺盛 ,连接呈网络 ;10d时细胞形态最为丰满 ;3周后细胞开始退化 ,胞内出现空泡。结论 培养的脊髓神经元形态发育符合正常规律变化 ,适用于形态学、生化、遗传、药理等学科研究
【Abstract】 Objective To find an easy,practical and reliable method for dispersion culture of primary neurons in rat spinal cord.Methods Spinal cord was collected from fetal or newly born Wistar rats,then with combination of trypsase digestion and mechanical trituration,tissues were dispersed into cell suspension which was cultured in 10% DMEM of fetal bovine serum and added cytarabine to inhibit reproduction of gliocytes for observation of cell growth.Results The morphologic development of cultured neurons was in conformity with regular pattern.Conclusion This method can be used in research about neuron morphology,biochemistry,genetics and pharmacology.
