【摘要】 激活素A是一种由卵泡液中提取出来的蛋白 ,主要以旁分泌和自分泌途径调节卵泡颗粒细胞功能 ,而对卵泡发育的影响却不十分清楚。本实验目的旨在探讨激活素A对大鼠离体培养卵泡发育的影响。用胶原酶分离窦前卵泡并分别加入激活素A(140 μg/L)、FSH(0 1IU/mL)以及卵泡抑素 (FS)在DMEM培养液中进行离体卵泡培养 4d ,观察卵泡直径、雌二醇分泌的变化。结果表明 :激活素A处理组和FSH +激活素处理组的卵泡直径显著增大 (P <0 0 1)。FSH +激活素A处理组雌二醇的分泌显著增加。FS可抑制激活素A对卵泡发育的影响。结果显示 :激活素A可以促进未成年大鼠窦前卵泡的生长发育更多还原

【Abstract】 Activin A is a kind of protein isolated from follicular fluid and is recognized as a local regulator of granulosa cell by paracrine or autocrine. Whether activin A promoted folliculogenesis still remains unelucidated. The aim of this experiment was to elucidate the effect of activin A on follicular growth in rat preantral follicle culture. Preantral follicles were isolated and cultured in DMEM medium supplemented with activin A(140μg/L),FSH(0.1IU/mL), and FS(100μg/L) for 4 days. Follicular diameter was measured daily and the amount of estradiol was determined by RIA.The results showed that Preantral follicles had a significantly increase in diameter when cultured with activin A or activin A plus FSH. Activin A and FSH treatment increased estradiol secretion by folicles from immature rat.The action of activin A was partially inhibited by follistatin.These results indicated that activin A promoted the growth of preantral follicle from immature rats.更多还原
