利用分子标记对桃属植物种的识别及其亲缘关系分析
The Identification and Systematic Relationship Analysis of Amygdalus spp. by Molecular Markers
【摘要】 采用RAPD技术 ,用筛选的 2 2个随机引物对桃属主要种类及其类型进行DNA扩增分析。结果表明 :不同引物扩增出的DNA片段范围在 3~ 13条 ,分子量范围在 30 0bp~ 3kb之间 ,依据特征谱带 ,可建立种间及其类型识别的分子检索表。利用 2 2个引物扩出的 180个位点上的RAPD带 ,进行聚类分析 ,结合前人研究出的结果 ,揭示桃种间亲缘演化顺序为 :内蒙古长柄扁桃→光核桃→山桃→甘肃桃→白花山桃、红花山桃、帚形山桃→桃巴旦、陕西桃巴旦→新疆桃、毛桃。
【Abstract】 Total DNAs of major species and types in Amygdalus L. were amplified with selected 22 arbitrary primers by using RAPD.The results showed that the number of DNA fragments amplified by different primers varied from 3 to 13,molecular sizes from 300 bp to 3 kb.The tested accessions could be identified by the amplified bands. According to special bands, a chart capable of distinguishing the species and types was figured out. The cluster and analysis of RAPD bands in 180 loci of 22 arbritrary primers, involving the relative research results, revealed the systematic evolution of the experimented species as follow:Neimenggu changbinbiantao( A. pedunculata )→Guanghetao( A.mira )→Shantao( A.davidiana )→Gansutao( A.kansuensis )→Baihuashantao,Honghuashantao,Zouxingshantao( A.davidiana )→Taobadan,Shanxi taobadan( A.communis )→Xinjiangtao( A.ferganensis ),Maotao( A.persica ).
