【摘要】 根据国外已发表的猪环状病毒 2型 (PCV 2 )的全基因组序列 ,设计一对 2型特异性引物 ,对可疑病料进行PCR扩增。将扩增产物连接到pMD 18 T载体上并克隆到大肠杆菌DH5α中 ,经提取质粒进行PCR、酶切、测序鉴定 ,证明扩增出了PCV 2的目的片段。将测序结果与GenBank收录的PCV 2序列进行比较 ,发现同源性均在90 %以上。用该PCR方法在 43份可疑病料中检测到了 12份PCV 2阳性病料 ,表明该病在我国已有流行更多还原

【Abstract】 According to the published genome sequences of porcine circovirus type 2 (PCV?2) a pair of specialprimers was designed to detect PCV?2 in samples from some farms by PCR. The PCR product was extracted and linked to pMD 18?T vector and then was colned into E.coli .DH5α.The recombination plasmid was identified by digestion with different enzyme restrictions, PCR and sequencing analysis. The homology of nucleotide sequence between the PCR product and other PCV?2 strains was more than 90%. Within forty three samples, twelve positive samples were found by PCR. It showed that PCV?2 infection is popular in our country.更多还原
