【摘要】 目的 :探讨胰岛素和转化生长因子 β(TGF β)对体外生长的人牙周膜 (PDL)细胞生物学特性的影响。 方法 :通过体外细胞培养技术、酶动力学法和考马斯亮蓝法 ,检测胰岛素和TGF β单独或联合应用时对人PDL细胞碱性磷酸酶 (ALPase)活性和总蛋白含量的影响。结果 :单独应用时 ,胰岛素浓度 1 0～ 10 0U/L ,TGF β浓度 0 1～ 10 0 μg/L ,明显促进人PDL细胞的ALPase活性和总蛋白含量 ,最佳浓度分别为 10U/L和 1μg/L ,以二者的最佳浓度联合应用时 ,作用更为显著 (P <0 0 1)。结论 :胰岛素和TGF β均能促进人PDL细胞的分化和总蛋白合成功能 ,二者联合应用有协同效应更多还原

【Abstract】 Objective:The aim of this study is to evaluate the biological effects of insulin and/or transforming growth factor (TGF_β) on alkaline phosphatase (ALPase) activity and protein synthesis of periodontal ligament (PDL) cells in vitro. Methods:The fifth passage of PDL cells were cultured in a 5% humid CO\-2 environment at 37℃ by using Dulbecco minimum essential medium (DMEM) supplemented with 10% fetal bovine serum (FBS) for 24 hours, and then insulin, TGF_β, and insulin and TGF_β were added respectively in a medium of 1% FBS. After 5 days, their ALPase activities were measured by using a kinetic method, and the total protein content was measured by using Coomassie brilliant blue staining. Results:Both insulin and TGF_β significantly increased the ALPase activity and the total protein content of PDL cells at the concentrations from 1.0 to 100U/L, and of 0.1～100μg/L. The most optimal concentrations of insulin and TGF_β were respectively 10U/L and 1μg/L. The synergistic effect of insulin (10 U/L) and TGF_β (1μg/L) was observed (P<0.01). Conclusion:The results suggest that insulin and/or TGF_β may promote the differentiation and protein synthesis of PDL cells .The combination of them may be valuable in the regeneration of periodontal tissue.更多还原
