1-己胺甲酰基-5-氟尿嘧啶的新合成路线
The New Synthetic Route of HCFU
【摘要】 本文提出了抗癌药物 1-己胺甲酰基 -5 -氟尿嘧啶不使用剧毒光气的新合成路线 :利用库尔提斯 (Curtius)重排反应从庚酸乙酯制备异氰酸己酯 ,然后再与 5 -氟尿嘧啶缩合得到产物。该合成路线操作条件温和、易于控制 ,且与环境友好。
【Abstract】 In this paper the new synthetic route of HCFU was based on the Curtius rearrangement reaction,in which HCFU was prepared by two steps. Firstly,the n -hexyl isocyanate was prepared from ethyl heptanoate and hydrzine hydrate by Curtius condensation reaction. And then the title compound was synthesized from 5-FU by using pyridine as solvent,in which the mixture of 0.1mol 5-Fu and 0.1mol n -hexyl isocyanate in pyridine was heated 3h at 95℃ to give HCFU.
