指状青霉诱发大鼠细胞DNA突变效应研究

【摘要】目的 :研究指状青霉 (Penicilliumdigitatum)的致突变效应。方法 :采用E .coliND 16 0菌株 ,大鼠肺及肝原代细胞非程序DNA合成 (UDS)试验和E .coliK12infA基因突变试验及infA基因序列测定和分析。结果 :指状青霉提取物 :①可明显地诱发E .coliND 16 0菌株回复突变 ;②可明显诱导大鼠肝原代细胞UDS((P <0 .0 5 ) ,极显著诱导肺原代细胞UDS(P <0 .0 1) ;③可诱导E .coliK12infA基因DNA序列中 5个碱基位点突变 ,且其中 1个位点的突变还可导致编码相应氨基酸的变异。结论 :指状青霉对原核细胞和真核细胞DNA均有明显的致突变性更多还原

【Abstract】 Aim:To study the mutagenicity of the extract from penicillium digitatum.Methods:Reverse mutation assay,unscheduled DNA synthesis (UDS) in the primary cells of the lung and liver of rat and E.coli K12 infA gene mutation assay(the sequences of mutated infA gene were determined and analyzed).Results:The extract could induce the reverse mutation of E.coli ND 160 strain and UDS in the primary cells of the lung and liver of rat( P <0.01 and P <0.05);Five points nucleotide mutation of infA gene were induced by the extract and one of them led to amino acid variation(Lys Val).Conclusion:It is indicated that penicillium digitatum have obvious effect on mutagenictiy.更多还原
