左旋肉碱对5-Fu所致大鼠肠粘膜屏障破坏的保护作用
The protective effects of L-carnitine on gut barrier injury caused by 5-Fu in rats
【摘要】 目的 :研究腹腔注射 5 Fu后左旋肉碱对大鼠肠粘膜形态及屏障功能的影响及其机制。方法 :36只SD大鼠随机分为空白组、对照组和实验组 ,每组各 12只 ,实验组和对照组均以 5 Fu腹腔灌注造成大鼠肠粘膜屏障破坏的模型 ,实验组在 5 Fu腹腔灌注前 4d给予左旋肉碱灌胃。分别观察实验第 1d ,4d ,7d的体重变化 ,第 8d处死后行细菌移位检测 ,常规病理切片观察肠粘膜厚度的变化 ,测定结肠及回肠组织中丙二醛 (MDA)含量及过氧化氢酶(CAT)、一氧化氮合酶 (NOS)的活性。结果 :①实验组中结肠及回肠的粘膜厚度均较对照组为高 ,但以结肠粘膜的厚度增加更显著 (P <0 .0 5 ) ;②对照组 2例细菌移位阳性 (2 / 12 ) ,空白组和实验组均未见细菌移位 ;③实验组回肠、结肠组织中CAT的酶活性较对照组均明显升高 (P <0 .0 5 ) ;实验组回肠、结肠组织中NOS酶活性变化及MDA含量则较对照组有所减少 ,但以结肠组织中的减少更明显 (P <0 .0 5 ) ;④第 7d对照组体重的减轻与空白组、实验组相比差异有显著性 (P <0 .0 1)。结论 :左旋肉碱对 5 Fu所致大鼠肠粘膜屏障的破坏有一定的保护作用 ,并可能参与改善化疗时的营养状况。
【Abstract】 Aim:To study the effect of L carnitine on intestinal mucosa morphology and gut barrier function in SD rats treated with 5 Fu introperitoneally Methods: Thirty six SD rats were randomly divided into 3 groups: blank, control and experiment groups. In both control and experiment groups 5 Fu was given introperitoneally to make the model of rat gut barrier injury, whereas in the experiment group carnitine was given ig 4 d before use of 5 Fu. Then the body weight change on the 1 st, 4 th and 7 day, was recorded the gut mucosal thickness was measured by histological section, and the level of malondialdehyde(MDA)and activity of catalase(CAT) and nitric oxide synthase(NOS) were tested in all groups. Results: ① Mucosal thickness of the colon and ileum in the experiment group was higher than that in the control group, but the increase in colon mucosa was significant ( P <0 05)compared to the control group. ② Two positive cases of bacterial translocation were only found in control group(2/12). ③ activity in the colon and ileum in experiment group was significantly higher than that in the control group( P <0.05), NOS activity and MDA level in experiment group reduced to some extent,with significant decrease in the colon tissue( P <0.05). ④ The levels of weight of the control group 7th on the day significantly decreased compared to those of blank and experiment groups( P <0.01).Conclusion: It is suggested that carnitine might play an important protective role in gut barrier injury in rats challenged by 5 Fu, and probably can improve the nutrition condition at chemotherapy
