酸性神经肽1对小鼠大脑环核苷酸含量的影响

【摘要】 目的 :探讨酸性神经肽 1(bovineacidicneuropeptide 1,BANP 1)对小鼠脑内环核苷酸含量的影响。方法 :将小鼠随机分为 6组 (每组 10只 ) ,其中 5组 (实验组 )按 15mg/kg腹腔注射BANP 1,将小鼠在注射后 0 5h ,1h ,2h ,3h ,4h处死并取脑匀浆 ,另一组为对照组 ,注射生理盐水 ,在注射生理盐水后 1h处死并取脑匀浆 ,用体积分数为75 %冰乙醇抽提环核苷酸类物质并沉淀蛋白 ,应用放射免疫法测定小鼠脑环磷酸腺苷 (cyclicmonophosphateadeno sine,cAMP)和环磷酸鸟苷 (cyclicmonophosphateguanosine ,cGMP)。结果 :腹腔注射BANP 1后 ,不同时间的结果分别与对照组比较 :cAMP结果差异无显著性 (P >0 0 5 ) ;cGMP浓度有一定升高 ,1h、2h、3h时显著升高 (P <0 0 5 ) ,其余各时间组差异无显著性 (P >0 0 5 )。结论 :BANP 1急性用药后在一定时间可升高脑内cGMP水平 ,对cAMP水平无显著影响。

【Abstract】 Aim:To probe the changes of cyclic nucleotides (cAMP,cGMP) in mouse’s brain caused by bovine acidic neuropeptide 1(BANP 1).Methods: Mice were divided into 6 groups (10 in each group) and 5 groups were injected with BANP 1 into the abdominal cavities at dose of 15 mg/kg,and at 0.5 h,1 h,2 h,3 h and 4 h the mice’s brains were collected and homogenated with 75% alcohol for cAMP and cGMP examination.The other group was injected with normal saline as control group and the mice’s brains were collected and homogenated at 1 h for examination.cAMP and cGMP were analysed by radiation immunity method.Results:Compared with that of control group,cAMP concentration had no significant difference at different moment ( P >0 05).cGMP concentration increased at 1 h,2 h and 3 h ( P <0 05).Conclusion:The injection of BANP 1 can increase the concentrations of cGMP at different time,but have no significant influence on cAMP concentration.
