【摘要】 目的 :观察妥泰 (TPM)单药对难治性癫痫 (RE)的临床疗效和安全性。方法 :RE4 1例 ,添加TPM治疗后逐渐撤去原服用的抗癫痫药物 ,而用TPM单药治疗。以添加TPM前 12周平均每 4周发作频率为基线 ,与评价前12周平均每 4周发作频率进行个体自身比较。按常规计算完全控制率和有效率。并进行临床观察和实验室检查。结果 :完全控制率 34 1% ,有效率 6 5 9%。不良反应 2 5例次 ,但绝大部分轻微 ,可自行消失。结论 :TPM对RE疗效显著 ,安全性好更多还原

【Abstract】 Objective:The efficacy and safety of topiramate(TPM)as adjunctive therapy and subsequently as monotherapy in the treatment of Chinese patients with refractory epilepsy were investigated in an open cilnical study.Methods:A total of 46 patients who had two or more seizures within a 4-week baseline phase were enrolled.TPM was titrated to target doses of 200mg/d for adults or 50mg～100mg/d for children for 8-week and maintained at stabilized levels for another 12 weeks.When patients were seizures-free or the target doses were reached,the doses of concomitant antiepileptic drugs(AEDs) gradually decreased,and AEDs were finally stopped in all patients.Clinical and laboratory monitoring were performed at baseline and after treatment with TPM.Results:In all,41 patients completed the trial.The proportion of patients with seizure-free was 34.1% and with a>or=50% reduction from baseline was 65.9%.Adverse events were mostly mild and transient.Routine blood cell counts and other laboratory results showed no significant changes from baseline.Conclusion:TPM 200mg/d for adults or 50mg～100mg/d for children is effective and well tolerated as treatment for refractory epilepsy.更多还原
