金鱼尾鳍组织的体外培养及血清对其生长的影响

【摘要】 采用组织块培养法对金鱼不同组织进行培养, 并研究了血清对细胞生长的影响. 在26℃温度下,采用添加15% 胎牛血清的199培养基成功地获得了金鱼尾鳍细胞系. 以2(105 个/ml的细胞密度接种时,细胞倍增时间为3.3 d, 金鱼尾鳍细胞培养系统中的最适血清浓度为15%.

【Abstract】 The present study was for providing experimental materials for virology, molecular biology and cell biology. Culture of cells from different tissues and the growth of cells in medium with different serum concentration were investigated. A cell line derived from caudal fin of goldfish, Carassius auratus, was developed successfully using explant techniques. The cell line grew well in Medium 199 supplied with 15% FBS at 26℃. The duplication time of the cell line was 3.3 d at a plating density of 2.0(105 cells/ml. It is proved that the cultures in Medium 199 supplied with 15% FBS showed the best result.
