血管紧张素Ⅱ受体Ⅰ基因A1166C多态性对Ⅱ型糖尿病患者发生肾病的影响
Effect of AT1 A1166C Gene Polymorphism on the Etiopathogenesis of Diabetic Nephropathy
【摘要】 目的 :探讨血管紧张素Ⅱ受体I(AT1 )的A116 6C基因多态性与糖尿病肾病的相关性 ;AT1 的A116 6C基因多态对Ⅱ型糖尿病肾病发病因素的影响。方法 :用聚合酶链反应 限制性片段长度多态性 (PCR RFLP)分析法探查AT1 的A116 6C基因多态性在健康组、单纯Ⅱ型糖尿病组 (以下简称DM组 )、糠尿病肾病组 (以下简称DN组 )的基因频率分布 ,并分析其对以上疾病发病因素的影响。结果 :各组之间并未发现明显不同的等位基因分布 ;各组内AT1基因型与各因素间的单因素偏相关分析表明 :DM组中AC基因型与空腹胰岛素水平 (FINS)和血清甘油三酯 (TG)呈正相关 (r1 =0 .42 9 ,r2 =0 .2 5 5 ) ;DN组中AC基因型与脂蛋白 (a)呈正相关 (r=0 .318 )。结论 :提示AT1 基因A116 6C多态与糖尿病肾病无关 ,但携带C等位基因的患者易于发生脂代谢紊乱
【Abstract】 Objective:To investigate whether the angiotensin Ⅱ receptor Ⅰ(AT 1)A1166C gene polymorphism is associated with diabetic nephropathy(DN) in type Ⅱ diabetes patients; To study the effect of AT 1 A1166C gene polymorphism on the pathogenesis of DN in type Ⅱ diabetes patients. Methods: We investigated the allele frequency and the genotype distribution of the AT 1 A1166C gene polymorphism in the group of normal control, simple diabetes, diabetic nephropathy by means of polymerase chain reaction restrictive fragment longevity polymorphism(PCR RFLP).Results: Genotype AC is positive associated with fast insulin(FINS) and triglycerides(TG) in diabetes group(r 1=0.429,r 2=0.255) and with lipoprotein (a) in DN group(r=0.318). Conclusion: There is no association between the AT 1 A1166C gene polymorphism and DN, but the diabetes and DN patients who bring a C allele of AT 1 gene are prone to develop lipid disorders.
