微生物酶拆分方法生产D-泛酸的手性中间体D-泛解酸内酯

【摘要】 筛选到一株产D -泛解酸内酯水解酶的串珠镰孢霉菌 (FusariummoniliformeSW -90 2 )。产酶条件研究表明 ,用甘油作碳源 ,蛋白胨作氮源 ,初始 pH8.0 ,温度 2 6℃ ,摇瓶培养 3d ,产酶量最高。在 6 0L和 10 0 0L发酵罐中通风发酵 4 5~ 4 7h ,产酶量为 6~ 8g干菌体 /L ,D -泛解酸内酯水解酶酶活力达到 0 .87~ 0 .92IU/g干菌体。该酶的最适反应温度为 5 5℃ ,最适反应 pH为 7.0~ 7.5。在酶不对称水解泛解酸内酯过程中 ,对溶液加酶量 5 %~ 10 % ,底物浓度 10 %~ 2 0 % ,控制水解率 2 0 %~ 30 % ,水解效果最好

【Abstract】 A strain of Fusarium moniliforme SW 902 for producing D lactonohydrolase was isolated and identified. Using glycerol as carbon source and peptone as nitrogen source, the maximum enzyme activity was obtained at 26℃ with initial pH 8.0. When the organism was cultured in the 60L or 1000L fermentor under the optimum conditions for 45~47h, 6~8g dry cells /L and 0.87~0.92 IU/g dry cell weight of specific activity were obtained. The conditions for enzyme enantioselective hydrolysis were also studied. The optimum temperature and the optimum pH for the enzyme reaction were 55℃ and pH 7.0~7.5, respectively. And with a substrate concentration of 5%~10% and enzyme concentration of 10%~20%, the maximum hydrolysis rate was obtained.
