三氧化二砷对肝癌裸鼠移植瘤模型作用的实验研究

【摘要】 目的 :研究三氧化二砷 (As2 O3)抑制肝癌细胞增殖、诱导肝癌细胞凋亡以及对骨髓抑制等作用。方法 :建立肝癌裸鼠移植瘤模型 ,应用 TU NEL 法观察不同浓度的 As2 O3对裸鼠移植瘤凋亡指数 (AI)的影响 ;应用免疫组化 SABC法检测不同浓度的 As2 O3对增殖细胞核抗原阳性细胞指数 (PI)的影响 ;检测移植瘤裸鼠的 WBC并进行骨髓涂片检查。结果 :经 As2 O3治疗的移植瘤裸鼠的骨髓有核细胞增生活跃 ,粒红比例正常 ,巨核细胞多见 ;低、中浓度 As2 O3治疗裸鼠移植瘤的 AI高于对照组 (P <0 .0 1和 P <0 .0 5 ) ;高浓度组 AI与对照组比较 ,无显著性差异 (P >0 .0 5 )。低、中浓度组 PI低于对照组 (P <0 .0 1) ;高浓度组 PI与对照组比较 ,无显著性差异 (P >0 .0 5 )。结论 :As2 O3通过抑制肿瘤细胞增殖、诱导细胞凋亡而达到其抗肝癌的效果 ,As2 O3对移植瘤裸鼠骨髓无抑制作用。

【Abstract】 Objective:To study the effects of arsenic trioxide (As 2O 3) on inhibiting the proliferation of hepatic carcinoma and inducing the apoptosis of hepatic carcinoma,and to find out whether or not it has the side effect of bone marrow inhibition Methods:To detect the apoptosis index (AI) and proliferation cell nuclear antigen (PCNA) positive nuclei cell index (PI) of the As 2O 3 treate nude mice transplants of hepatic carcinoma by using the TUNEL detection and SABC method,and to detect the side effect of bone marrow inhibition of As 2O 3 by counting the peripheral blood WBC and by bone marrow smear Results:As 2O 3 did not have the side effect of bone marrow inhibition AI was higher (P<0 01 or P<0 05),PI was lower (P<0 01) in low and middle concentration As 2O 3 groups comparing with the control group Conclusions:The anti-tumor effects of As 2O 3 on hepatic carcinoma were inhibiting proliferation and inducing apoptosis of tumor cells As 2O 3 did not have the side effect of bone marrow inhibition
