柑橘酸性转化酶基因片段的克隆

【摘要】 分析植物液泡和细胞壁转化酶基因的保守区序列,分别设计一对PCR引物,以柑橘基因组DNA为模板,采用PCR方法分别扩增出长约740 bp和530 bp的DNA片段,分别克隆入pBS-T 和pMD18-T载体,进行测序,结果表明,获得柑橘酸性转化酶基因家族的两个成员,成员A和B基因片段分别长742 bp和524 bp。其序列已在GenBank中登记(登记号分别为AF014925和AF314197)。在GenBank中进行同源性检索,结果表明,成员A编码的氨基酸与植物液泡酸性转化酶的氨基酸同源性较高,与拟南芥的最高,达76%,而成员B编码的氨基酸与植物细胞壁酸性转化酶的氨基酸同源性较高,与豌豆的最高,达71%。两成员核苷酸和氨基酸序列同源性分别为55% 和50%,推测酸性转化酶基因成员A 和B编码的蛋白分别定位于液泡和细胞壁。

【Abstract】 Two pairs of primers, designed respectively from conserved regions of plant vacuolar and cell wall acid invertases, were used to amplify 742 bp and 524 bp fragments by polymerase chain reaction (PCR). The fragments were cloned into pBS-T and pMD18-T, respectively, and then sequenced. It is suggested that two members (member A and B) of citrus acid invertase gene family were obtained. The deduced amino acid sequence of member A is 76% identical to the gene from Arabidopsis thaliana (GenBank AC005698), and was suggested to encode a soluble acid invertase located in vacuole, while member B is 71% identical to the gene from garden pea (Pisum sativum, GenBank X85327), and was suggested to encode a insoluble acid invertase located in cell wall.
