芯片毛细管电泳-激光诱导荧光-电荷耦合器件检测系统
Microchip-Based Capillary Electrophoresis with Laser Induced Fluorescence-Charge Coupled Device Detection
【摘要】 采用自组建的芯片毛细管电泳 -激光诱导荧光 -电荷耦合器件 ( CCD)检测系统在数十秒内满意地分离了曙红和荧光素。设计了一种进样、分离电路 ,可以有效地消除进样通道的样品溶液向分离通道的渗漏。解决了由这种渗漏所引起的电泳峰变宽、拖尾等问题。提高了芯片毛细管电泳的分辨率和分离效率
【Abstract】 The microchip based capillary electrophoresis is blooming in recent years as a high speed and high efficient separation method. In this paper, EOS and fluorescein were separated in a few seconds by using setup of capillary electrophoretic microchip, laser induced fluorescence and charge coupled device made in our laboratory. In our experiment, a type of electrocircuit for sampling and separation was employed to prevent sample solution from bleeding into separation channel from sampling channel, which can cause electrophoretic peaks broaden and tailing. The resolution and separation efficiency were improved greatly by using this electrocircuit.
