衍生化毛细管气相色谱法分析血清中的游离脂肪酸
Analysis of Free Fatty Acids in Human Serum by Derivatization Capillary Gas Chromatography
【摘要】 用H2 SO4 甲醇对血清中游离脂肪酸进行甲酯化反应 ,考察了不同温度和时间以及不同的衍生条件对甲酯化完全程度的影响 ,确定了对血清中游离脂肪酸衍生化完全的条件为 :在 6 2℃下用 10 %H2 SO4 甲醇溶液衍生化 2h。大部分脂肪酸的衍生反应和色谱分析的相对标准偏差都在 3.0 0 %以内。用各脂肪酸和内标色谱峰面积的比值对脂肪酸浓度作标准曲线 ,线性回归系数在 0 .999~ 1之间 ,定量测定了 5个不同地区的 2 5 0个血清样品中 12种游离脂肪酸的含量
【Abstract】 The free fatty acids in human serum were esterified using H 2SO 4/methanol. The influence of temperature, derivatising time and different derivatizing conditions on the extent of esterification reaction was investigated. The free fatty acids in human serum can be esterified in 10% H 2SO 4/methnol at 62℃ for 2 h. The RSD for both derivatizing reaction and chromatographic analysis of most fatty acids was <3.00%. The standard calibration curve was obtained from the ratio value of chromatographic area of fatty acids and internal standard and the concentration of fatty acids. The linear calibration coefficients were between 0.999 and 1.000. 12 free fatty acids of 250 human serum collecting from five districts were quantitatively determined.
