龙眼胚性愈伤组织长期继代培养及其染色体数目变异
Long-term subculture of embryogenic calli and their genetic variation of the chromosome number in longan
【摘要】 从龙眼幼胚培养诱导并筛选出的松散型胚性愈伤组织 ,在继代培养基上已培养 6 a以上 ,所保持的若干愈伤组织仍具有强烈的体细胞胚胎发生能力 .经细胞学检查 ,其中个别愈伤组织系的部分愈伤组织团发现有2 n=3x=4 5,或 2 n=4 x=6 0等染色体数目的变异细胞 ,这对于离体筛选龙眼细胞突变体很有价值
【Abstract】 Friable embryogenic calli induced and then selected from the culture of longan ( Dimocarpus longan Lour.) immature embryos maintained strong capacity of somatic embryogenesis after having been subcultured for over 6 years. The cytological examinations showed that there were some cell variations with the chromosome number 2n=3X=45, or 2n=4X=60 or others in some cell clusters of callus lines, which was of great value for obtaining cell variants in vitro in longan.
