机油降解菌的筛选及其降解能力的研究
Screening of Lubrication Oil Degrading Bacterium Strains and Their Degradation Capability
【摘要】 从环境微生物工程角度出发,以20~#机油为唯一碳源,从含油污泥中筛选出3株降解机油能力较强的苗株,借助形态学观察和生理生化试验,鉴定为:动胶菌属(Zoogloea sp.)、氮单胞菌属(Azomonas sp.)和假单胞菌属(Pseudomonassp.).对上述3菌株分别进行摇瓶试验,在此基础上,选择除油能力最强的动胶菌属 LD_2菌株,进行生物接触氧化法处理含20~#机油废水的室内动态模拟试验.结果表明,动胶苗属LD_2 N株降解机油能力比较显著:当入流质量浓度为 424~1432 mg/L时,出流水中 20~#机油含量降为 0~20 mg/L,除油率达到97 %~100%.用18~#机油代替20~#机油,出流质量浓度降到4~10mg/L,除油率达到98%以上.以上经过处理的出流水质均达到国家污水排放标准.
【Abstract】 In order to treat wastewater containing lubrication oil by degrading bacteria, lubrication oil No. 20 was used as the sole carbon source to isolate and select bacterium genera which have high degrading capacity for lubrication oil in sludge contaminated with oil. Through morphological and biochemical methods, three types of bacterium genera were primarily identified as Zoogloea sp., Azomonas sp. and Pseudomonas sp. The Zoogloea strain showed the high- est capacity to degrade lubrication oil No. 20 among the tested strains in shaking flasks. On the basis of the preliminary results, Zoogloea strain was chosen to treat the wastewater contaminated by lubrication oil No. 20, using biological contacted oxidation method in small-scale tests. The wastewater was treated in three oxidation pools for different Hy- draulic Retention time( HRT) for 6 h and 12 h, respectively. The experiments showed a positive result that the con- centration of lubrication oil No. 20 was significantly reduced to 0-20 mg/L in the treated water, compared to the con- centration of 424-1 432 mg/L in the untreated water. The degrading rates varied between 97%-100%, and as a con- sequence, the quality of the treated water reached to the national standards for contaminated water. Lubrication oil No. 18 was also included in the test to examine the degrading capacity of Zoogloea strain. Similar results were obtained in this test, where the degrading rates for lubrication oil No. 18 was 98%, reaching to the national standards for contam-inated water.
