儿童及少年青春发育期血中高碱性磷酸酶活性的研究
A Study on High Serum Alkaline Phosphatase(AKP) Activity in Childhood and Puberty.
【摘要】 【目的】 对儿童及青春发育期血中碱性磷酸酶 (AKP)活性进行系统分析 ,初步探讨其分子特性及释放机制。 【方法】 收集儿童及青少年不同年龄组正常人血清 5 0 0例 ,进行酶活性分析、电泳、免疫测定、层析及酶分解试验。 【结果】 4~ 16岁儿童及青少年骨AKP活性超过 10 0IU/L。亲水型及疏水型均存在 ,2 0岁时降至正常 ,疏水型AKP不再存在。经磷脂酶D消化后 ,AKP由疏水型变为亲水型。了解到疏水型AKP其分子结构特点和释放机制。 【结论】 儿童及青少年期血清中骨AKP活性升高与成骨细胞活性密切相关 ,而肝AKP不随年龄而变化
【Abstract】 Objective With a systematic analysis of serum AKP in childhood and puberty to study the molecular character and releasing mechanism of AKP. Methods To analyse enzyme activity,electrophoresis,immunological measurment,chramatography and phosphatidase D of 500 normal child. Results The AKP activity in bone of childred aged 4~16 years was more than 100IU/L. It had two types:Hydrophilic and hydrophobic type.Their activity were down to normal when reached 20 years old and the hydrophobic type was no longer exit.When digested by phosphatidase D,the hydrophobic type can be changed to hydrophilic type.We got a understanding of the molecular configuration of hydrophobic type and the releasing mechanism of AKP. Conclusions There was close relationship between high serum AKP activity in bone and the activity of osteoblast.The AKP in liver was not changed by age.
