SDS聚丙烯酰胺凝胶电泳快速染色新方法的研究

【摘要】 通过几种金属盐溶液对 SDS聚丙烯酰胺凝胶电泳染色的实验表明 ,0 .2 5 mol/L的Ca Cl2 和 Mg Cl2 溶液能够对蛋白质进行有效的染色 ,经这 2种溶液染色的蛋白质都能够从凝胶中洗脱回收。尤其是 Ca Cl2 法灵敏度更高 ,而且蛋白质条带形成之后也十分稳定。所以在运用制备电泳纯化蛋白质时这种新的染色方法较适用

【Abstract】 Solutions of several metal salts were examined for their ability to stain polypeptides separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE).Protein patterns could be visualized less than 5 min after treatment of gels with calcium chloride.It was considered as a new rapid method for the detection of protein bands both in analytical and preparative SDS PAGE.Magnesium chloride can also stain proteins in SDS PAGE gel.However,it was less sensitive than calcium staining.Proteins from gels treated with the two new staining procedure could be easily recovered by electric elution.
