无根根霉脂肪酶的酶学性质研究及酯合成的应用
Studies on the character of rhizupus arrhizus fischer lipase and application in ester synthesis
【摘要】 初步探讨了无根根霉脂肪酶 (RhizopusarrhizusFischerLipase)的酶学性质及酯合成应用。结果表明 :在不同表现形式下该酶具有不同的最适pH、pH稳定性、最适温度和热稳定性。当用胆盐处理发酵液 ,Tween - 80处理菌体细胞时发现该脂肪酶活力分别提高 3倍和 3 5倍。本文利用溶致液晶固定化的方法在有机介质中催化酯合成 ,固定化后该酶酯合成的反应速度比其游离态高 30倍以上。最适条件下 ,合成油酸 - 2 -辛基十二烷酯的能力达 5× 10 - 5mol·hr- 1·u- 1。
【Abstract】 A strain of mold Rh.ar.Fischer producing lipase has been discussed.The character of RAF lipase and the use in estersynthesis were researched including optimum pH,stability of pH,optimum temperature and stability of heat.When dealt with culture media by cholate and handle mycelia by Tween-80,it showed hydrolysis activity of RAF lipase was increased by 300% and 350% respectively. A new method of lyotropic liquid crystallization with for the immobilization of lipase from RAF was used to catalyze ester synthesis in organic media.The speed of esterfication increased by about 30 times compared with that of its free one.The value was 5×10 -5 mol/hr·u.
