吉林大豆中卵磷脂的成分提取与分析

【摘要】 对大豆中卵磷脂的提取方法做了系统的研究 .采用混合溶剂(V正己烷∶V乙醇 =6∶4 )作为大豆中油提取溶剂的方法 ,出油率可达 1 7.6%~2 0 .8% ,与文献上报道的 1 6.8%有明显提高 ,大豆中的卵磷脂得到有效的分离提纯 .详细研究了薄层色谱法中不同极性的展开溶剂的影响 ,采用V氯仿∶V甲醇∶V水 =65∶2 2 .5∶4 (pH =4 .0 )做展开溶剂 ,经显色后 ,对薄层色谱板做光密度扫描 .结果表明 ,可对大豆中的磷脂酰胆碱 (PC)和磷脂酰乙醇铵 (PE)进行有效的分离 .该方法用于 1 1种吉林大豆中卵磷脂的成分分析 ,取得了满意的结果 .

【Abstract】 Method of extracting lecithin from soybeans and the analysis of lecithin by thin-layer chromatography were studied in the present paper. A method of using mixture solvent (hexane-ethanol-water) as extracting solvent was proposed, and the yields of oil extraction were 17.6%~20.8 %, higher than that of 16.8 % reported in literature. The effects of disperse solvents with different polarities and the pH on separation of lecithin on thin-layer plates were studied in details. Mixture of chloroform-methanol-water was used as disperse solvent and scanning thin-layer chromatograms were obtained. Results showed that PC and PE components can be separated and analyzed with the method proposed. This method was applied to extraction and analysis of lecithin in eleven soybean samples in Jilin province of China
