红皮云杉胚乳中提取DNA的最佳时段

【摘要】 以来自不同地点的红皮云杉胚乳为材料提取DNA研究了DNA提取的最佳时段.从而为构建红皮云杉的遗传图谱做准备.结果表明:提取DNA样品的最佳时段是在种子生根的晚期、即根长到4cm以后.提取的 DNA样品纯度和浓度都较高;单一胚乳中DNA量完全能够满足大量 PCR扩增的需要。

【Abstract】 In this paper. it mainly approaches the best time to extract DNA from endosperms of Picea koraiensis Nabal. which will be the starting work of making the genetic map for the conifer species in the future. The results are shown as follows: it is feasible to extract total DNA from single endosperm; the best time to extract DNA is in the late time of seed germination. with the root in 4cm length or more. when the nutrition in seed lins been used out. and also the amount and concentration of extracted DNA are all higher than that in the early time. Although one endosperm is small. extracted DNA is enough to meet PCR amplification reactions.
