【摘要】 利用 RAPD技术对美洲狼尾草不育系 2 3A、2 3DA、85 DA,恢复系 3B- 6及其相应的 3个F1杂交种基因组 DNA进行了多态性分析 ,摸索出一套适合美洲狼尾草 DNA提取、PCR扩增方法。对随机选取的 2 7个引物进行扩增 ,结果有 5个引物扩增出多态性条带 ,其中引物S369扩增的条带不仅重复性好 ,而且清晰 ,为美洲狼尾草亲本材料及其杂交种鉴定和纯度分析提供了一种快速、准确的检测手段。更多还原

【Abstract】 The genomic DNA for cytoplasmic male-sterile lines（23A, 23DA, 85DA）, a R-line（3B-6） and their F₁ hybrids （23A/3B-6, 23DA/3B-6,85DA/3B-6） of Pennisetum americanum were studied by means of RAPD. A suitable method about extraction and amplification of genomic DNA was established. Among 27 random primers, polymorphic bands were amplified from S 351,S 415,S 369,S 484,S 489 primers and the band by S 369 was of good replication and clearity. A rapid and correct method can be supplied to identify and analyze the purity of Pennisetum americanum.更多还原
