根癌农杆菌介导茶树转化研究

【摘要】 通过检测GUS瞬间表达 ,对影响根癌农杆菌侵染茶树叶片效果的若干因素进行研究。Kan对茶树叶片愈伤组织形成的抑制浓度在 3 0mg/L左右 ,因而对以Kan为筛选标记的载体来说 ,5 0mg/LKan是较为适宜的选择浓度。茶树较为适宜的农杆菌转化系统是用OD6 0 0 为 0 5 - 0 8的LBA4 4 0 4侵染 10分钟左右 ,暗中2 8℃共培养 2 - 3天。硝酸银对农杆菌生长有抑制作用 ,不宜在共培养阶段添加

【Abstract】 Some factors which affect the result of Agrobacterium tumefaciens infection leaf of tea plant (Camellia sinensis) was studied based on GUS transient expression 50 mg/L Kanamycin is suitable selection concentration for expression vectors with neomycin phosphotransferase gene (NPTⅡ) as callus formation from tea plant leaf was depressed by about 30 mg/L Kanamycin The suitable system of Agrobacterium tumefaciens _mediated transformation is suggested that tea plant leaf was infected 10 min by LBA4404 of OD600 about 0 5-0 8 followed by co_culture of 2-3 days about 28℃ in darkness. AgNO3 may inhibit the growth of Agrobacterium tumefaciens, so is not suitable to be applied into co-culture medium.
