黑曲霉U_γ-2(Aspergillus niger U_γ-2)产菊粉酶的研究

【摘要】 研究了黑曲霉Uγ- 2的产酶进程及其产酶条件。结果表明 :在 1g·mL-1菊芋汁中添加 30mg·mL-1的牛肉膏 ,调节初始pH 6 0 ,摇瓶转速为 180r·min-1,30℃条件下发酵最高酶活力为 180 33μmol·min-1·mL-1。发酵初期产酶速率较快 ,pH下降速度快。在发酵进行到 72~ 96h期间 ,出现产酶的迟缓期 ,此时发酵液的pH为 4 6 2~ 4 38;以后菊粉酶活力明显上升。在 192h时菊粉酶活力达到最高值 ,发酵液pH降到最低点。在发酵进程中 ,发酵液可溶性蛋白出现 2个高峰 ,72h时出现第一高峰 ,与菊粉酶活力比较表明此时主要是杂蛋白产生所致 ;在 192h时出现的第二高峰与菊粉酶活力同步 ,说明主要是菊粉酶产生。与生物量的变化曲线比较 ,尽管在发酵初期产酶速率较快 ,但菊粉酶在发酵液中大量积累出现在 12 0h后 (对数增长期的后期 )

【Abstract】 The process and controlling conditions of inulinases yielding from Aspergillus niger U γ-2 were studied The maximal inulinase activity(IA) was 180 33 μmol·min -1 ·mL -1 , with the culture of 1g·mL -1 Jerusalem artichoke juice(JAJ) in addition of 30 mg·mL -1 beef extract, at the conditions of initial pH 6 0, shaking speed 180 r·min -1 and 30℃ The rate of inulinase yielding and that of pH declined were fast in the initial fermentation phrase The maximum of IA and the minimum of pH showed at the same time (192 h) There are two soluble protein peaks occurred in the fermentation course The first one was given at 72 h, which was composed of the impure proteins, and the other showed the synchronism and the increase of IA Analysis of the changes of biomass(BM) showed that the optimal inulinase yielding phase was the period of 120~192 h (anaphase of the logarithmic growth phase and the whole stationary phase)
