【摘要】 用无血清培养系统研究了牛腔前卵泡的体外培养。直径为 1 0 0～ 2 0 0 μm的牛腔前卵泡在添加L -谷氨酸、BSA、睾酮、转铁蛋白和硒的McCoy’s 5a培养液中培养 ,卵泡保持正常的形态结构并持续生长。培养的第 1 0d左右形成卵泡腔 ,成腔率约50 %。培养液中添加胰岛素 ,对卵泡直径的增长和卵泡腔的形成有明显的促进作用 ,但添加FSH对腔前卵泡的生长未表现促进作用更多还原

【Abstract】 A serum-free culture system was used to study the in vitro culture of the preantral follicles from bovine ovaries. Follicles with diameters of 100～200μm, dissected mechanically from ovarian cortical slices, were cultured individually in 96-well plates with 250μL McCoy’s 5a medium supplemented with 20mM Hepes, 3mM L-glutamine, 0.1%BSA, 29ng/ml testosterone, 2.5μg/ml transferrin and 4ng/ml selenium. The follicles were incubated in a humidified atmosphere with 5%CO 2 at 37℃. The culture medium was changed every 6 days. The results showed that the follicles grew continuously maintaining there normal morphology. Antrum formation occurred after about 10 days of culture. The addition of insulin to the culture medium stimulated the growth of preantral follicles. However, similar effect was not shown for the addition of FSH.更多还原
