显微注射对绵羊体外受精胚胎的影响
Development of Sheep IVF Embryos After DNA Pronuclear Injection
【摘要】 本文研究了显微注射对绵羊体外受精胚胎发育的影响。显微注射组和对照组的囊胚率分别为 9.1%和 32 .2 % (P <0 .0 5 ) ;利用慢速冷冻的方法 ,显微注射组和对照组的囊胚的冷冻解冻后的存活率没有差异 (分别为 75 .5 %和 77.7% ,P >0 .0 5 )。显微注射组和对照组的冷冻胚胎移植后的产羔率分别为 2 5 .0 %和 33.3% (P >0 .0 5 )。结果表明 ,绵羊合子原核显微注射外源DNA后的发育能力下降 ,而发育到囊胚阶段的胚胎的抗冻性及移植后的发育能力受到的影响较小
【Abstract】 Development ability of sheep embryos after pronuclear injected with DNA was studied. The proportion of DNA injected zygotes developing into blastocyst was lower significantly than that of control group (9.1% and 32.2%, respectively, P<0.05). the survival rate of DNA injected blastocysts were similar to that of control group (75.5% and 77.7%,respectively, P>0.05 ), After transfer the thawed embryos, there is no significant difference in lamb rate between two groups (25.0% and 33.3%, respectively, P>0.05). In conclusion, this study show the development ability of sheep embryos decreased after DNA pronuclear injection. But blastocysts derived from DNA pronuclear injection can be cryopreserved successfully.
